چکیده
Recent reports identified DGAT1 (EC 2.3.1.20) harboring a lysine to alanine substitution (K232A) as a candidate gene with a strong effect on milk production traits. Our objective was to estimate the frequency of the DGAT1 K232A polymorphism in the main Zebu and Taurine breeds in Brazil as well as in Zebu x Taurine crossbreds as a potential QTL for marker-assisted selection. Samples of 331 animals from the main Brazilian breeds, Nellore, Guzerat, Red Sindhi, Gyr, Holstein, and Gyr x Holstein F1 were genotyped for DGAT1 K232A polymorphism (A and K alleles) using the PCR-RFLP technique. The highest frequency of the A allele was found in the Holstein sample (73%) followed by Gyr x Holstein F1 (39%). Gyr and Red Sindhi showed low frequencies of A alleles (4 and 2.5%, respectively). The A allele was not found in the Nellore and Guzerat samples. Our results could be used to guide association studies between this locus and milk traits in these breeds.

Dgat1 Polymorphism K232a in Sahiwal (indian Zebu) and Frieswal (holstein Friesian X Sahiwal Crossbred) Cattle
The positional candidate gene DGAT1, within QTL of BTA14, harboring a lysine to alanine substitution (K232A) is significantly associated with milk production traits. The present investigation was undertaken to estimate the frequency of the DGAT1 K232A polymorphism in Indian zebu breed, Sahiwal (B. indicus; N= 51) and crossbred, Frieswal (HF X Sahiwal; N= 126) cattle using PCR-RFLP technique. متن کاملDGAT1 and ABCG2 polymorphism in Indian cattle (Bos indicus) and buffalo (Bubalus bubalis) breeds
BACKGROUND Indian cattle (Bos indicus) and riverine buffalo (Bubalus bubalis) give a poor yield of milk but it has a high fat and protein percentage compared to taurine cattle. The identification of QTLs (Quantitative Trait Loci) on BTA14 and BTA6 and its subsequent fine mapping has led to identification of two non conservative mutations affecting milk production and composition. Our objective ...
متن کاملEffects of DGAT1 variants on milk production traits in German cattle breeds.
Various QTL mapping experiments led to the detection of a QTL in the centromeric region of cattle chromosome 14 that had a major effect on the fat content of milk. Recently, the gene encoding diacylglycerol O-acyltransferase (DGAT1) was proposed to be a positional and functional candidate for this trait. This study investigated the effects of a nonconservative lysine to alanine (K232A) substitu...

This experiment was aimed to study the association between the DGAT1 K232A polymorphism and milk production traits and somatic cell score (SCS) in Iranian Holstein dairy cows. The records of 408 animals from five dairy herds were randomly identified and then genomic DNA was extracted from blood using the modified-salting method described by Miller. RFLP-PCR was performed to obtain all the polym...
